Technological Breakthroughs and Innovations

This standard establishes for the first time a multiple nucleotide polymorphism (MNP) identification system, which achieves interspecies identification through 22 specific marker sites. Compared with traditional physiological and biochemical methods:

  • The detection cycle is shortened from 7 days to 3 days
  • The identification accuracy is increased to 99.7%
  • The detection limit is up to 10 CFU/g

Key Operation Points

Specifications for Culture Medium Configuration

Photosynthetic bacteria isolation agar needs to be added with 5 mL of ascorbic acid supplement (Appendix A.2), and the temperature needs to be kept below 46°C when pouring the plate. The double-layer plate method can effectively inhibit the growth of miscellaneous bacteria.

Molecular identification quality control

MNP detection requirements:
1. DNA purity A260/280 ≥ 1.7
2. Sequencing coverage ≥ 700X
3. Detection of ≥ 3 marker sites can be considered positive


Implementation suggestions

  1. Anaerobic environment must be maintained during sampling, and it is recommended to use pre-reduced phosphate buffer
  2. The colony spread samples should be pretreated by the membrane filter method
  3. Establish a standard strain quality control library and regularly verify the sensitivity of the reagents
