Interpretation of the core content of the standard

This standard specifies a standardized method for determining the antibacterial activity of polypeptide substances using the Oxford cup diffusion method, and calculates the antibacterial potency (AU/mg) by measuring the diameter of the inhibition zone.


In-depth analysis of technical principles

Based on the diffusion characteristics of polypeptides in agar plates: the effective antibacterial components diffuse from the Oxford cup to the surrounding culture medium, forming a concentration gradient, and a clear inhibition zone is generated when the local concentration is higher than the minimum inhibitory concentration.

Quality assurance measures

  • Use a vernier caliper (precision 0.02mm) for three-point measurement
  • Set up 3 parallel replicates for each concentration
  • Standard strain ATCC25923 is used as a quality control reference

Recommendations for the implementation of the standard

The laboratory should establish a standard strain library and regularly verify the temperature uniformity (±1℃) of the constant temperature incubator. It is recommended to use an automated inhibition zone measuring instrument to improve data reproducibility.
