Introduction

Technical Background of the Standard

Martei disease is an important parasitic disease of bivalve molluscs and is listed by OIE as a reportable aquatic animal disease. This standard replaces part of SC/T7207.3-2007 and standardizes the microscope tissue method as an independent diagnostic method for the first time, solving the technical connection problem between on-site rapid detection and laboratory diagnosis.

Key technical points

1. Sample processing specifications

Differentiate sampling according to the growth stage of shellfish: Visceral mass is taken from juvenile shellfish, Stomach/intestine/digestive gland/adductor muscle needs to be separated from adult shellfish. DAFA fixative (ethanol-formalin-acetic acid system) must be prepared and used immediately, and the fixation time should not exceed 24 hours.

2. Microscopic diagnostic features

The development of refractory Maltese shows obvious stages:
Larva stage: 4-8μm in diameter, mostly distributed in the gastric epithelium
Cyst stage: 20-40μm in diameter, concentrated in the digestive gland
Diagnostic structure: refringent bodies are the key identification feature


Implementation recommendations

  1. Seasonal prevention and control: Strengthen monitoring when the water temperature is >17℃, and increase the sampling frequency during the high temperature period in summer
  2. Result cross-validation: It is recommended that positive samples be reviewed in combination with the SC/T7207.3-2007 electron microscopy method
  3. Biosafety: Live shellfish from the epidemic area need to be isolated and observed to prevent the introduction of type O/M strains

Technology Evolution Analysis

This standard has major improvements over the 2007 version:
1. Added Mayer-Bennett Hematoxylin staining formula (Appendix A.2)
2. Clarified the thresholds for determining parasites at different developmental stages
3. Added molecular biological confirmation requirements for suspected cases in non-endemic areas
