Using double-labeled PCR technology:

  1. The 5' end of the upstream primer is labeled with FITC fluorescein
  2. The 5' end of the downstream primer is labeled with biotin
  3. The amplification product is detected by immunochromatography test strips:
    • Gold-labeled anti-FITC antibody captures the fluorescent labeled end
    • Fixed streptavidin captures the biotin labeled end
