Technology evolution analysis

Compared with the 2014 version of the standard, the main upgrades of the 2020 version are:

  1. Added Liquid chromatography-tandem mass spectrometry as the third method, with a 40% reduction in detection limit
  2. Optimized the sensitivity expression method and uniformly adopted the limit of quantification (LOQ) indicator
  3. Expanded the applicable sample types to special matrices such as Chinese medicinal materials

Key operating points

Sample pretreatment

Uniformly adopt the boiling water extraction method (95℃ water bath for 10min), note:

  • TCM samples need to pass through a 40-mesh sieve (pore size 0.425mm)
  • Tea is prepared according to GB/T 8303
  • The extract needs to be filtered through a 0.45μm aqueous filter membrane

Method selection recommendations

Case:When a tea company tested theanine:

  • Method 1 is used for routine quality inspection (low operating cost)
  • Method 3 is recommended for export testing (high international recognition)
  • Method 2 is used for scientific research (GABA and other special amino acids can be detected simultaneously)

Implementation precautions

  1. Derivatization reagents should be prepared and used immediately (especially AQC reagents)
  2. The pH of the ammonium formate mobile phase needs to be calibrated regularly for mass spectrometry (3.0±0.1)
  3. The reaction column temperature of the amino acid analyzer needs to be checked daily (135±0.5℃)
  4. Cystine needs to be tested during method validation (easy to lose by oxidation)
