Technology evolution analysis
Compared with the 2014 version of the standard, the main upgrades of the 2020 version are:
- Added Liquid chromatography-tandem mass spectrometry as the third method, with a 40% reduction in detection limit
- Optimized the sensitivity expression method and uniformly adopted the limit of quantification (LOQ) indicator
- Expanded the applicable sample types to special matrices such as Chinese medicinal materials
Key operating points
Sample pretreatment
Uniformly adopt the boiling water extraction method (95℃ water bath for 10min), note:
- TCM samples need to pass through a 40-mesh sieve (pore size 0.425mm)
- Tea is prepared according to GB/T 8303
- The extract needs to be filtered through a 0.45μm aqueous filter membrane
Method selection recommendations
Case:When a tea company tested theanine:
- Method 1 is used for routine quality inspection (low operating cost)
- Method 3 is recommended for export testing (high international recognition)
- Method 2 is used for scientific research (GABA and other special amino acids can be detected simultaneously)
Implementation precautions
- Derivatization reagents should be prepared and used immediately (especially AQC reagents)
- The pH of the ammonium formate mobile phase needs to be calibrated regularly for mass spectrometry (3.0±0.1)
- The reaction column temperature of the amino acid analyzer needs to be checked daily (135±0.5℃)
- Cystine needs to be tested during method validation (easy to lose by oxidation)
