Introduction

Interpretation of standard technology

Technical elements Characteristics of this method Comparison with traditional methods
Detection limit Carbadox 5.0μg/kg, MQCA 1.0μg/kg ELISA method 10-50μg/kg
Pretreatment time About 2 hours 4-6 hours
Specificity Double confirmation in MRM mode High antibody cross-reactivity rate

Key technological innovations

The Oasis MAX solid phase extraction column combined with the Tris buffer system (pH 9.6) was used for purification, which significantly improved the removal rate of fat-soluble impurities. In the optimization of mass spectrometry parameters, carbadox selected the parent ion m/z 263.2→231.2, and MQCA selected m/z 175.0→102.1 as the quantitative ion pair.

Key points for implementation

  1. The sample needs to be enzymatically hydrolyzed with proteinase K at 45℃ for 2 hours
  2. 0.1% formic acid-acetonitrile (9:1) is used as the extraction solvent
  3. The chromatographic column is C18 (100mm×2.1mm, 2.7μm)

Standard Evolution Analysis

Compared with GB/T 20746-2006, this standard adds:

  • Simultaneous detection of metabolite MQCA
  • Improved matrix-matched calibration curve method
  • Expanded to feed matrix (the original standard is limited to animal tissue)
