Background of Standard Formulation and Analysis of Technical Evolution
Glutamine transaminase is an important biochemical reagent widely used in food processing, pharmaceutical manufacturing and other fields. In order to ensure its activity and consistency in industrial products, the national standard GB/T 34795—2017 came into being. This standard was proposed and managed by the National Technical Committee for Biochemical Testing Standardization. The main drafting units include Shenzhen Institute of Metrology and Quality Inspection, etc.
Definition of glutamine transaminase activity unit
According to the definition in standard 3.1, the glutamine transaminase activity unit is the amount of enzyme required to catalyze the production of 1.0 μmol of isohydroxy acid salt per minute at 37°C and pH 6.0. This definition provides a clear quantitative basis for the detection method.
Detection Principle
Transglutaminase catalyzes N-benzyloxycarbonyl-L-glutamylglycine (CBZ-Gln-Gly) and hydroxylamine to generate L-glutamic acid-Y-monoisohydroxy acid. After the product is colored with ferric chloride, it shows a characteristic absorption peak at a wavelength of 525nm, so the enzyme activity can be calculated by the external standard method.
