《GB/T 35538-2017工业用酶制剂测定技术导则》由TC64(全国食品工业标准化技术委员会)归口,TC64SC5(全国食品工业标准化技术委员会工业发酵分会)执行,主管部门为国家标准化管理委员会。
Introduction
Standard Framework and Technology Evolution
GB/T 35538-2017 is my country's first national standard for industrial enzyme preparation assay technology. It integrates the technical requirements of existing industry standards such as QB/T 1803, and introduces the internationally accepted enzyme activity assay verification system. The standard took three years to formulate and was led by the China Food Fermentation Industry Research Institute and completed jointly by leading domestic and foreign companies such as Novozymes.
Analysis of core assay elements
| Key parameters | Selection principles | Typical range | Example |
|---|---|---|---|
| Substrate | Minimum Km value/strong specificity | 10-100 times of Km | Casein (protease) |
| Temperature | ±10℃ gradient test | 35-60℃ | Animal enzyme 35-40℃ |
| pH | Binding stability curve | 4.5-7.5 | Pullulanase pH4.5 |
| Buffer system | Within the range of pKa±1 | 0.05-0.2mol/L | Acetic acid-sodium acetate system |
Enzyme activity determination standardization process
Critical control points
- Standard curve verification: R²≥0.98, set 5-7 gradient points
- Control enzyme requirements: Contaminated enzyme needs to be detected by ELISA (<50,000ng/g)
- Precision control: Repeatability RSD≤4-10%
Comparison of typical methods
| Method type | Principle | Applicable enzymes | Precision requirements |
|---|---|---|---|
| DNS method | Reducing sugar color development | Pullulanase | CV≤12% |
| UV method | Substrate absorbance change | Catalase | CV≤8% |
| Dyeing substrate method | Color product precipitation | Cellulase | CV≤10% |
Implementation difficulties and solutions
Common problems
- Matrix interference: Acarbose is recommended to inhibit the activity of impurities
- Temperature drift: A constant temperature water bath with an accuracy of ±0.1℃ is required
- Substrate batch differences: 24-sample cross-validation is required
Method validation requirements
Need to meet the following requirements
- Accuracy 95-105% (referenced to standard substances)
- Intermediate precision ≤10%
- Linear range ≥2.5 orders of magnitude
Industrial application connection
The standard places special emphasis on the correlation verification between enzyme activity indicators and actual application performance. A supplementary testing plan should be established when the following situations occur:
- Synergistic/antagonistic effects exist in the complex enzyme system
- The difference between industrial reaction conditions and test conditions is greater than 15%
- The product contains auxiliary materials that affect the determination
It is recommended to use SDS-PAGE electrophoresis combined with HPLC for impurity monitoring.
