Standard Framework and Technology Evolution

GB/T 35538-2017 is my country's first national standard for industrial enzyme preparation assay technology. It integrates the technical requirements of existing industry standards such as QB/T 1803, and introduces the internationally accepted enzyme activity assay verification system. The standard took three years to formulate and was led by the China Food Fermentation Industry Research Institute and completed jointly by leading domestic and foreign companies such as Novozymes.

Implementation difficulties and solutions

Common problems

  • Matrix interference: Acarbose is recommended to inhibit the activity of impurities
  • Temperature drift: A constant temperature water bath with an accuracy of ±0.1℃ is required
  • Substrate batch differences: 24-sample cross-validation is required

Industrial application connection

The standard places special emphasis on the correlation verification between enzyme activity indicators and actual application performance. A supplementary testing plan should be established when the following situations occur:

  1. Synergistic/antagonistic effects exist in the complex enzyme system
  2. The difference between industrial reaction conditions and test conditions is greater than 15%
  3. The product contains auxiliary materials that affect the determination

It is recommended to use SDS-PAGE electrophoresis combined with HPLC for impurity monitoring.
