Introduction

Core Value and Application Scenarios of the Standard

This standard aims at the risk control needs of pesticide residues in the grain circulation link, and establishes a rapid screening system based on fluorescence detector. Compared with the traditional chromatographic method, the detection time is shortened from 4-6 hours to 30 minutes, which is particularly suitable for on-site rapid screening in the grain storage and processing links.


In-depth Analysis of Technical Principles

The detection is based on the enzyme inhibition method: organophosphorus/carbamate pesticides inhibit the activity of insect esterases, resulting in a decrease in the hydrolysis rate of fluorescent substrates.

Key operating points

Case study: When a grain depot tested dichlorvos residues in rice, it was found that the following operational differences had an impact on the results:

  1. When the incubation temperature was 35℃±1℃, the inhibition rate deviation was ≤5%
  2. Adding the fluorescent substrate for more than 30 seconds would cause the signal to attenuate by 15-20%
  3. When the sample size did not reach the 20-mesh standard sieve, the extraction efficiency decreased by 30%

Implementation suggestions and precautions

1. Matrix difference compensation: Different grain varieties should refer to Appendix C to select the corresponding limit enzyme inhibition rate. For example, the dichlorvos limit in brown rice is 45.6%, while that in corn is 50.6%

2. False positive processing: For suspected positive samples (inhibition rate exceeds the standard), GC-MS/MS should be used for confirmation in combination with the limit requirements of GB2763

3. Equipment calibration: Fluorescence detector needs to be verified monthly with standard products to ensure the accuracy of the 460-550nm excitation wavelength and 590nm emission wavelength
