Detailed explanation of the method principle
Based on a three-step color reaction:
1. MBTH reacts with reducing sugars to form azine under alkaline conditions
2. Fe3+ oxidizes excess MBTH to form cations
3. The cyan-blue compound produces characteristic absorption at 590nm
Key control points: 80℃ water bath for 13min requires precise control, and the color should be measured immediately after 1h of development.
Implementation recommendations
- The standard solution should be prepared and used immediately. The MBTH reagent is valid for only 10 days.
- The temperature deviation needs to be calibrated when using a constant temperature water bath.
- After the sample is dissolved, it needs to be left to stand and the supernatant should be taken to avoid turbidity interference.
- It is recommended to carry a blank control and a quality control sample for each batch of measurements.
