Analysis of the core content of the standard

This standard uses a spectrophotometer to determine the reducing end sugar content at the end of the water-soluble chitosan molecule, and establishes a quantitative relationship at 590nm through the MBTH-Fe3+ color development system.

Detailed explanation of the method principle

Based on a three-step color reaction:
1. MBTH reacts with reducing sugars to form azine under alkaline conditions
2. Fe3+ oxidizes excess MBTH to form cations
3. The cyan-blue compound produces characteristic absorption at 590nm

Key control points: 80℃ water bath for 13min requires precise control, and the color should be measured immediately after 1h of development.


Implementation recommendations

  1. The standard solution should be prepared and used immediately. The MBTH reagent is valid for only 10 days.
  2. The temperature deviation needs to be calibrated when using a constant temperature water bath.
  3. After the sample is dissolved, it needs to be left to stand and the supernatant should be taken to avoid turbidity interference.
  4. It is recommended to carry a blank control and a quality control sample for each batch of measurements.
