Key Operation Points

Sample Pretreatment: Weigh 2.5g of sample and extract it in a 40-50℃ water bath, then centrifuge at 12000r/min for 20 minutes and filter through a 0.45μm filter. Add isotope internal standard D6-inositol for calibration.

Chromatographic Conditions: The mobile phase is 0.2% formic acid aqueous solution-methanol (2+98), the flow rate is 0.20mL/min, the column temperature is 40℃, and the injection volume is 10μL.

  • Ion pair: Inositol m/z 179.1→161.0 (quantitative ion), 185.0→166.9 (internal standard ion)
  • Collision energy: Optimization in the range of -15V to -20V
  • Ion source parameters: ESI negative ion mode, nebulizer gas (N2) pressure 50psi

Implementation suggestions

  1. The standard solution should be prepared and used immediately and stored at 4℃ for no more than 3 days
  2. Blank test and spike recovery should be carried out simultaneously for each batch of samples
  3. It is recommended to use a triple quadrupole mass spectrometer such as AB Sciex 4000 QTRAP
