- Gel liquefaction phenomenon: Count the non-liquefied area to estimate the total amount
- Too dense colonies: Select representative grids for estimation (30 grid system)
- Edge diffusion: Use 3M pressure plate to ensure uniform distribution of sample liquid
Interpretation of verification data
According to the verification results in Appendix A:
- Linear relationship: Correlation coefficient R²>0.98 (8 matrices including pickled pepper chicken feet)
- Precision: RSD≤3.5% (high and low concentration samples)
- Density resistance: stable results within the range of 35-37℃, 45-51h
Implementation suggestions
- Establish test piece acceptance criteria: verify batch number consistency (see Appendix A.3)
- Complex samples need to be supplemented with GB 4789.35
- Periodically compare methods: it is recommended to perform a traditional plate method control once a quarter
