Key technical points

Innovation in sample pretreatment: The composite purification strategy of PSA adsorbent and n-hexane defatting is used to effectively solve the complex matrix problems of animal-derived samples. For example, honey samples need to be melted in a 60℃ water bath before testing to ensure homogeneity.

Mass spectrometry parameter optimization: m/z 749.5→591.3 was selected as the quantitative ion pair and azithromycin-D5 (m/z 754.5) was used as the internal standard to compensate for the signal suppression caused by the matrix effect (average correction efficiency 92-107%).


Implementation and application recommendations

  1. Quality control:Each batch of samples should be accompanied by blank controls and spike recovery experiments, and the recovery rate should be in the range of 80-110%
  2. Instrument maintenance:It is recommended to clean the ESI ion source after every 50 injections to prevent matrix deposition
  3. Method validation:When changing the brand of chromatographic column, the mobile phase gradient needs to be re-optimized (see standard Table 1)

Standard evolution analysis

Compared with the ELISA method of SN/T 2316-2009, the LC-MS/MS technology of this standard increases the specificity to 99.7% and reduces the false positive rate to 0.3%. The 2021 version adds validation data for royal jelly and casing matrices, expanding the applicability of the standard.
