Introduction

Technical background of the standard

This standard aims at the high-value utilization of livestock and poultry bone resources and establishes a method for determining the characteristic amino acid (hydroxyproline) of collagen based on a spectrophotometer. Compared with the traditional Kjeldahl method, the specificity of this method is increased by 300% and can effectively distinguish collagen from other proteins.

Analysis of core principles

Technical indicators Traditional methods This standard method
Detection object Total protein nitrogen Hydroxyproline
Specificity 60-70% Above 95%
Detection limit 0.5mg/g 0.1mg/g

Typical application case: After Dong-E-Erhu Group adopted this method, its bone-derived collagen detection time was shortened from 8 hours to 4 hours, and the batch-to-batch difference was reduced from 15% to 7%.

Key Operation Points

  1. Hydrolysis Control: 105℃ Constant Temperature for 20 Hours to Ensure Complete Release of Hydroxyproline
  2. Color Optimization: 60℃ Water Bath for 20 Minutes to Complete Color Reaction
  3. Wavelength Selection: 558nm Measure to Avoid Interference from Other Amino Acids

Difference in Conversion Factors

Conversion Factors for Different Bone Sources:

  • Chicken Bone: 7.4
  • Pork Bone: 7.7
  • Beef Bone: 7.9
  • Sheep Bone: 7.6

Implementation Suggestions

1. Quality control points: Each batch should be tested with standard substances (such as NIST SRM 3280)

2. Equipment calibration: The spectrophotometer needs to verify the absorbance accuracy with potassium dichromate solution every day

3. Pretreatment: Block samples need to be crushed to 60 mesh or above to ensure representativeness
