Sample Pretreatment Differences
For high-fat samples (such as ice cream and milk powder), a polyamide solid-phase extraction cartridge is required for cleanup. The eluent is 5% methanol in water and the elution is 2% ammoniated methanol. Due to the higher sensitivity of the LC-MS/MS method, the sample volume can be reduced to 2.5mL.
Mass spectrometry parameter optimization
LC-MS/MS was performed in multiple reaction monitoring (MRM) mode, and the quantitative ion pair for CurⅠ was 369.0→177.2 (CE=45eV). It should be noted that the DP voltage (51V) and collision energy needed to be re-optimized for different instruments.
Implementation Recommendations
- HPLC is the preferred method for routine screening, as it is low-cost and meets most testing needs.
- For samples with low limit requirements, such as infant food, LC-MS/MS must be used.
- Note that the reference materials should be stored in the dark (0-4°C). The CAS numbers of the three curcumin standards are shown in Appendix A.
- For complex matrices, it is recommended to add matrix-matched standard curves to reduce ion suppression.
Background of Standard Evolution
This standard was published in 2017. Compared to the earlier GB/T 22244-2008, the main improvements include: ① the addition of LC-MS/MS confirmation methods; ② expansion to 13 food matrices; and ③ optimization of the solid-phase extraction process. Even after the 2020 amendment to Regulation (EU) No 1129/2011 by the European Union, this standard remains technologically advanced.
